That is surely possible, however it would not be very pleasing.
The themer must add the following to browser.css:
toolbar:hover[iconsize="small"] #back-button {
list-style-image: url("chrome://browser/skin/large.png");
-moz-image-region: rect(48px 48px 96px 0px);
}
toolbar:hover[iconsize="small"] #back-button[disabled="true"] {
-moz-image-region: rect(48px 96px 96px 48px);
}
(for each toolbar button).
Notice the 'toolbar:hover[iconsize="small"]' which activates when 'hovering' over a
toolbar with small icons. One could also define the default icons for 'large' a small, and enlarge them
when hovering.
The effect is that when the mouse hovers the toolbar all icons become bigger,
however the 'browser' pane itself shrinks, causing a reflow of the page, which is
annoying.

You can make each icon grow, on hover, within the confines of the toolbar height (I think)- say 22px to 24px -maybe 32 to 40px - would that work for you? The new Marvilla brights direction icons change size, slightly, and form on hover. You would have a slightly larger toolbar, for the icons, to start with though - to allow for growth.

I can't - I don't know if it's possible (unless you run your CPU at 100%! - LOL!). Growth would be instant
Sure it's possible. Probably pretty easy by using flash to style the buttons
