Was something changed in Nightly to cause incompatibility with Decentraleyes?

Maybe, it's also occurring on http://gpu.userbenchmark.com/Compare/AM ... 924vsm7753 which didn't happen a week or so ago. I haven't got around to testing yet though.

you should get " restore defaults " an " Exceptions " buttons do you get either one?

same here, clicking dont change anything, also defaults or exceptions (which are empty). anyhow its nightly...

I get both buttons.
In 09-02 build, I think the feature can be enabled (and functional), even though there is no ✓ in the checkbox. The sub-options, i.e. "only in private windows" and "always", are now selectable.

Strange, I can toggle it here....

EDIT: I had not noticed I was NOT running latest Nightly, after update I too cannot enable Content Blocking and appears that tracking protection is also affected, i.e. NOT enabled
