I see that TB has an option to let you enable and select the digital certificate to use for signing and encrypting email.
However, when I try to select a certificate I get a message that the certificate manager could not locate a valid certificate.
Where is the TB certificate manager looking for the certificate and how do I get a certificate loaded there?

Opening the thunderbird certificate manager
To open the certificate manager and add digital certificates, run the following on the command line (Win32):
thunderbird.exe -chrome chrome://pippki/content/certManager.xul
I believe the same command line options should work on other platforms as well.

One thing
One thing to note.. Thunderbird must already be open for the previous command to work.
