If you are running Windows 7, Right click the executable for firefox and select run as Admin. the video will then play. Easy work around until Adobe gets its act together.

5. Go to C:\Documents and Settings\[Your User]\Application Data\Mozilla\Firefox\Profiles
6. Copy the file permissions.sqlite from the new profile folder to the original profile folder
7. Start Firefox
This worked in style...

I was only seeing this issue in Firefox, not in Chrome.
Disable the browser feature that allows the user to save content to Real Player:

Frank Lion is right. My situation is that I had to replace my hard drive which I had had for several years in mid June 2012 and reinstall all software from scratch. Alas most Youtube videos wouldn't load, giving "error, try back later" sorts of messages. I had loaded the latest version (11) of flashplayer when I replaced the drive. So I uninstalled flash in the control panel and then went to the Adobe site and downloaded an obsolete version of flashplayer "fp_10.3.183.20_archive.zip" and like magic, Youtube works perfectly once again.

But in RealPlayer Firefox is not shown as a browser which is set-up for downloading.
Only Internet Explorer and Chrome are!
So how would deselecting that affect Firefox?
In any case I want to be able to download with RealPlayer in IE and Chrome

Don´t know, but affect Firefox it did. Tried all other suggestions and nothing worked. Lost all saved data though due to not reading last page first

hea, im not sure if our cause for the error is the same... but i know mine was called by an out of date real player flash download plugin... its frustrating... but wen i turn the extension off... my youtube video starts working again.

I've had the YouTube site telling me (in the vid box) "An error occurred. Please try again later."
But...omg you've saved me a ton of trouble!
I DISABLED all of my "Shockwave Flash" plugins (just the "Shockwave Flash" plugins)/ Closed the browser / Then reopened the browser and went straight to YouTube, and now ALL the vids play! (I don't know exactly how--maybe I have another plugin like "shockwave flash" but under a different name? I wouldn't know.) Again THANKS!!

Problem solved as new version of QuickTime installed
In my case of Mozilla 14.0.1 on Win7 the problem disappeared after I updated the QuickTime plugin. As I understood, it was rather an installation of a newer version than an update because I had to download QuickTimeInstaller.exe (37 MB) and install it over the existent one.
I suggest to update all add-ons/extensions: press Ctrl+Shift+A to reach the list, click sth like "check if the extensions are not outdated" [English version may differ because I translate from Ukrainian], apply all available updates (you will be redirected to the extension developers' websites).
Luckily, the problem disappeared without any experiments with cookies.
