Thunderbird is offering an update from version 45.8.0 to 52.0.1 but says it will disable the calendar extension.
So is Lightning dead?

No. That type of build problem occasionally happens. Sometimes what you need to do is uninstall the existing version of Lightning and get the latest from the Mozilla add-ons web site. I didn't notice anything about it in the release notes or whats new.
A quick search found https://bbs.archlinux.org/viewtopic.php?id=224903&p=2 which states "52.0-2 is still built without the calendar feature but it now works with the Lightning add-on 5.4b4, which 52.0-1 did not." and "Just removed the latest stable release of Lightening, upgraded to 52.0-2. Following the upgrade installed lightening 5.4b4 and the calendar is back in operation." They're referring to a archLinux package so its possible 52.0-2 is equivalent to 52.0.1 from Mozilla.
Personally, I stick with the less popular ReminderFox add-on instead since my needs are simple and its a mature add-on that is unaffected by these type of build problems.

No, it indicates it will be disabled, then when 45.8.0 updates, Lightning updates with it.
At least it did, when I did update testing of 45.8.0 to the 52.0 and 52.0.1 release candidates.

Thank you, it updated and the calendar extension is still there. I love Thunderbird.
