Nope, mine's been extremely slow to start for probably over a year or two now with uBlock Origin enabled (on the first launch of the day anyways). Some suggest wiping my profile and/or refresh, but I just don't want to do it.

But I did just find that FF/Nightly's Autoplay blocking makes nbcnews' current/new video player completely unusable, even when it's just set to mute.

That link works fine for me when I allow video and audio autoplay, and then reload the page.

Yes, that's pretty much why I said the autoplay blocker makes it unusable when active.

Could this be related to the uBlock advanced function that prevents tabs from loading until uBlock is ready?

You could try switching that off to see if it helps, bur bear in mind that if you do this, some network requests are likely to get past uBlock on initial startup.
